I think the poison is to protect her, by building immunity. He thinks of her as useful for his goal. Much easier to control, without any loyalty to his enemy. I think he wants her to be his spy, or lover, or some other useful tool.

I know there’s lots of chapters to read, but my theory is that there’s no novel.
Lots of people have been brainwashed by the prince, to believe that they’re heroes in a story that puts him in a favorable light and the duke is presented as a monster. Maybe he wasn’t such a ‘madman’ but the legion of brainwashed people trying to get him must have tired him.
Maybe he saw that Susan was at least not a devoted fan to the prince and spared her (I supposed that the others were ‘I prefer to die than to serve you!’). Maybe that’s why Susan seems to have ‘spawn’ suddenly three years ago.
Maybe the ‘poison’ is something to break the brainwashing ? Idk
I also bet that the noble girl in the annex (Ariel ?) is also trying to escape from the prince and the people that try to make a couple of them ‘like in the novel’.
Just a theory, but for me it’s the only way that the duke could be excused for his behavior.
